Modification of late human embryo development after blastomere removal on day 3 for preimplantation genetic testing

In this retrospective single center study, time-lapse monitoring of embryo development was used to assess if embryo (blastomere) biopsy on day 3 after fertilization, for preimplantation genetic testing, had an effect on the average time specific morphological changes of the embryo (morphokinetics). In total the development of 4269 embryos was tracked in this study (1691 biopsied embryos, 2578 non-biopsied embryos) in a cultured time-lapse monitoring system.

Early morphokenitics, up to day 3, between both groups was similar. Significantly several morphokenitic parameters after day 3 biopsy significantly differed between the 2 groups. This included the onset of compaction, fully-compacted morula stage, onset of cavitation/early blastulation, and blastocyst stages (tB, tEB) which occurred significantly earlier in the biopsy group.

Limitations

  1. This study does not suggest modification to the embryos genetic expression
  2. The biopsy procedure was not strict, with either 1 or 2 blastomeres extracted, per embryo
  3. The effect of embryo biopsy on implantation rate was not evaluated
  4. There are significant differences between the 2 groups regarding ovarian reserve and smoking status, which could influence results
